JPMorgan maintains "Neutral" rating on Johnson & Johnson (JNJ.US): Strong data from two anti-cancer drugs, but valuation premium limits upside potential

Johnson & Johnson’s new prostate cancer drug and head and neck cancer drug Ervantumab both released positive data at ESMO. JPMorgan stated that the pipeline shows differentiated potential but valuations are relatively high, maintaining a Neutral rating.

According to Golden Ten Data APP, the European Society for Medical Oncology (ESMO) annual meeting has just concluded. This is one of the world's most important academic conferences in the field of oncology, and the data released at the event often directly impacts the stock prices and pipeline expectations of relevant pharmaceutical companies. Johnson & Johnson (JNJ.US) became one of the most closely watched large-cap pharmaceutical stocks. In a research report released on October 25, JPMorgan assessed that early data from Johnson & Johnson in two major indications—prostate cancer and head and neck cancer—provides potential differentiated competitiveness for its innovative drug pipeline.

The core highlight lies in JNJ-6143, a prostate cancer candidate drug, with the technology platform named Halda Riptac. This is an investigational therapy targeting metastatic castration-resistant prostate cancer (mCRPC). This indication usually refers to patients whose disease progresses after receiving androgen receptor pathway inhibitors, representing a late-stage population with unmet clinical needs. The phase I clinical data this time is quite impressive: enrolled patients were at a highly advanced stage, with the median patient previously receiving four lines of systemic therapy, and over 30% had been treated with taxane chemotherapy and radioligand therapy.

In the selected 50 mg cohort advancing to phase II, the objective response rate (ORR) reached 75%, and after a median follow-up of 5.6 months, 71% of patients were still receiving treatment, with efficacy unaffected by genomic characteristics. Even more crucial was the safety profile: in the 50 mg cohort, the incidence of grade 3 or higher treatment-related adverse events was zero.

Analyst Chris Schott believes this data suggests that JNJ-6143 may have a differentiated advantage in both efficacy and tolerability, receiving high recognition from on-site discussion panelists as well. Based on this, Johnson & Johnson will advance the drug to phase III clinical trials and simultaneously expand to metastatic castration-sensitive prostate cancer and combination therapy directions. Analysts expect that by the 2030s, the vast prostate cancer market could provide substantial incremental growth for the company.

Another main focus is head and neck cancer. The OrigAMI-4 trial cohort five for Rybrevant (amivantamab) in combination with Keytruda (pembrolizumab) and chemotherapy demonstrated that, among HPV (human papillomavirus) negative head and neck cancer patients, the combination's objective response rate reached 77% (sample size: 22), significantly higher than the approximately 36% response rate with pembrolizumab plus chemotherapy in this indication. Johnson & Johnson has already submitted a marketing application for the second-line head and neck cancer indication for Rybrevant, while first-line phase III clinical trials are also progressing. The report indicates that this drug is expected to occupy an important position in the field of HPV-negative head and neck cancer.

Nevertheless, the report's view on Johnson & Johnson’s stock remains relatively restrained. Analysts pointed out that Johnson & Johnson is still a fundamentally sound company among large pharmaceutical firms, with its innovative drug business leading with high single-digit growth and its medical technology business migrating to high-growth market segments.

JPMorgan maintains a "Neutral" rating on Johnson & Johnson, with a latest target price of $285. Based on estimated 2027 earnings per share, the company’s valuation has reached 21 times earnings, which is clearly at a premium level. Relative to other comparable companies, there is limited upside potential for the stock price, which also explains why analysts remain cautious on the share price despite optimism on the fundamentals.
